Hygromycin B is an aminoglycoside antibiotic produced by the bacterium Streptomyces hygroscopicus that kills bacteria, fungi and higher eukaryotic cells by inhibiting protein synthesis.
Hygromycin has been used to select drug-resistant stable transfectants after transfer of the hygromycin phosphotransferase gene in a variety of cell culture applications. It is used in gene transfer experiments as a selection agent.
Mode of action: Inhibition of protein synthesis. It interferes with translocation of the 70S ribosome by inducing the misreading of the m-RNA template in prokaryotes and eukaryotes. It selectively penetrates cells that have been rendered permeable by virus infection.
Hygromycin B is provided as a caramel-coloured solution in water. It is not prepared under sterile conditions. If a sterile solution is desired, working solutions of Hygromycin B in aqueous media can be passed through a 0.2 micron filter before use.
Freely soluble in water, methanol, buffer solution and ethanol. Practically insoluble in less polar solvents.
