Sulpho-NHS reacts primarily with primary amines to convert carboxyl groups to amine reactive sulfo-NHS esters. It is ideal for cross-linking, chemical labelling and solid support immobilisation. It is used to generate amine reactive esters from carboxylate groups and is primarily used to increase the efficiency of the carbodiimide EDC cross-linking. EDC reactions do not require Sulpho-NHS; however, its presence greatly enhances cross-linking efficiency and allows for a two-step process. EDC reacts with a carboxylate group to form an active ester, which is subject to rapid hydrolysis in aqueous buffers. The Sulfo-NHS increases the stability of the active intermediate and the resulting coupled product is identical to EDC reactions without sulpho-NHS.
Sulpho-NHS is water-soluble, long-lived, and hydrolyses relatively slowly in water and in aqueous buffer, unlike NHS.
