SfiI restriction enzyme recognizes GGCCNNNN^NGGCC sites and cuts best at 50 °C in G buffer.

Storage buffer: 10 mM Tris-HCl (pH 7,4 at 25 °C), 300 mM NaCl, 10 mM MgCl2, 1 mM DTT, 0,15% triton X-100, 0,2 mg/ml BSA and 50% (v/v) glycerol. After 50-fold overdigestion with SfiI, more than 95% of the DNA fragmentscan be ligated and recut. Minimum 5 units of theenzyme are required forcomplete digestion of 1 µg of agarose-embedded adenovirus-2 DNA in 16 hours.
Applications include molecular cloning, restriction site mapping, genotyping, southern blotting, restriction fragment length polymorphism (RFLP) and SNP.
