β-Secretase (Beta-site amyloid precursor protein (APP) cleaving enzyme 1/BACE1), participates in the initial cleavage of amyloid precursor-APP generating two soluble N-terminal and C-terminal fragments. Another secretase isozyme, γ-Secretase, cleaves C-terminal fragment producing amyloid β.
Accumulation of amyloid β in brain results in the formation of amyloid plaques, which contribute in the development of Alzheimer’s disease (AD) in humans. Therefore, screening for novel and specific inhibitors of human β-Secretase (BACE1) has become critical in Alzheimer's disease-related drug discovery research.
Human β-Secretase (BACE1) inhibitor screening kit, human BACE1 cleaves a quenched substrate, specific for BACE1, generating a product with high fluorescence that can be measured at 435/500 nm. In the presence of BACE1 Inhibitor (control included in this kit), the enzymatic cleavage is impeded and could be used to screen for new inhibitor compounds.
