IL-7 supports the growth of early B cells from long-term lymphoid bone marrow cultures. It is mitogenic to thymocytes and enhances the response of cells to other stimuli such as polyhydroxyalkanoate (PHA) and concanavalin A (ConA). IL-7 stimulates the proliferation of CD4+/CD8+ cells.

There are at least four distinct but related alkaline phosphatases in humans: intestinal AP (IAP), placental AP(PLAP), germ cell AP (GCAP) and their genes are clustered on chromosome 2, tissue-nonspecific isozyme (TNAP) which gene is located on chromosome 1. Alkaline phosphatases (APs) are dimeric enzymes, it catalyse the hydrolysis of phosphomonoesters with release of inorganic phosphate. The native ALPL is a glycosylated homodimer attached to the membrane through a GPI-anchor. This isozyme may play a role in skeletal mineralisation. Mutations in ALPL gene have been linked directly to different forms of hypophosphatasia,characterised by poorly mineralised cartilage and bones, and this disorder can vary depending on the specific mutation since this determines age of onset and severity of symptoms.

In contrast to the B7 family members which contain two IgG domains, VSIG4 contains one complete V-type Ig domain and a truncated C-type I g domain. VSIG4 is abundantly expressed in several fetal tissues. In adult tissues, the highest expression of VSIG4 is in lung and placenta. It is also expressed in resting macrophages. No VSIG4 expression appears to be present in T and B cells. The specific expression of VSIG4 on resting macrophages in tissue suggests that this inhibitory ligand may be important for the maintenance of T cell unresponsiveness in healthy tissues. VSIG4 functions as a negative regulator of T cell activation, and may be involved in the maintenance of peripheral T cell tolerance, and is also identified as a potent suppressor of established inflammation. VSIG4 is a phagocytic receptor, strong negative regulator of T-cell proliferation and IL2 production. It is a potent inhibitor of the alternative complement pathway convertases. Human VSIG4 is 399 amino acids (aa) in length. It is a type I transmembrane (TM) glycoprotein that contains a 264 aa extracellular domain (ECD) (aa 20 - 283) and a 95 aa cytoplasmic region.

Each monomer of Transthyretin has two 4-stranded beta sheets and the shape of a prolate ellipsoid. Antiparallel beta-sheet interactions link monomers into dimers. A short loop from each monomer forms the main dimer-dimer interaction. These two pairs of loops separate the opposed, convex beta-sheets of the dimers to form an internal channel. Defects in Transthyretin are the cause of amyloidosis type 1 (AMYL1) which is a hereditary generalized amyloidosis due to transthyretin amyloid deposition. Protein fibrils can form in different tissues leading to amyloid polyneuropathies, amyloidotic cardiomyopathy, carpal tunnel syndrome, systemic senile amyloidosis. The disease includes leptomeningeal amyloidosis that is characterized by primary involvement of the central nervous system.
