Beschreibung: Osteopontin (OPN) is an acidic extracellular matrix cell adhesion protein that is relatively abundant not only in bone matrix, plasma, urine, and milk, but is also found in malignant and atherogenic tissues. Phosphorylation, glycosylation and calcium modifications allow intact and fragmented Osteopontin to direct a variety of diverse responses including tissue remodeling, inflammation and cell survival. Plasma Osteopontin has been shown to provide prognostic information in breast, prostate, colon, and lung cancers as well as metastatic carcinomas. The notable presence of Osteopontin in a variety of tumors is strongly correlated to pathological stage, suggesting its critical role in tumor invasiveness, progression and metastasis. In addition, Osteopontin inhibits inducible nitric oxide synthase activity, thereby protecting tumor cells from NO-mediated macrophage cytotoxic attack. Osteopontin is found in atherosclerotic plaques and may drive a number of diabetic vascular pathologies.

Beschreibung: All members of the inhibitor of apoptosis proteins (IAP) family contain at least one, but usually three copies of baculovirus IAP repeat (BIR), a ~70-aa zinc binding domain, and upon overexpression suppress apoptosis. The BIR motif is capable of interacting with caspases and occluding their catalytic pockets. Certain IAPs also possess C-terminal RING domains. RING-containing proteins often act as E3 ubiquitin ligases and can catalyse the degradation of both, themselves and selected target proteins through ubiquitylation. So far, eight human IAPs have been identified: Apollon (Bruce), c-IAP1 (HIAP2; MIHB), c-IAP2 (HIAP1; MIHC), ILP-2 (Ts-IAP), Livin (ML-IAP; KIAP), NAIP, Survivin (TIAP) and XIAP (ILP-1; MIHA). c-IAP1 has been shown to inhibit specific caspases.

Beschreibung: The ryanodine receptor (RyR) is the channel responsible for the release of calcium from the sarcoplasmic reticulum (SR) in muscle cells and also plays a role in calcium regulation in non-muscle cells. The RyR exists as a homotetramer and is predicted to have a short cytoplasmic C-terminus and 4-10 transmembrane domains; the remainder of the protein, termed the "foot" region is located in the cytoplasm between the T-tubule and the SR. The mammalian RyR is the product of three different genes: RyR-1, which is expressed predominantly in skeletal muscle and areas of the brain, RyR-2, which is expressed predominantly in the heart muscle but also found in the stomach, endothelial cells and diffuse areas of the brain, and RyR-3 which is found in smooth muscle and the brain (striatum, thalamus and hippocampus).
