Beschreibung: Programmed death-ligand 1 (PD-L1), also known as CD274 or B7-H1, is a protein that in humans is encoded by the CD274 gene. It is mapped to 9p24.1. PD-L1 is a 40kDa type 1 transmembrane protein that has been speculated to play a major role in suppressing the immune system during particular events such as pregnancy, tissue allografts, autoimmune disease and other disease states such as hepatitis. It has been concluded that upregulation of PD-L1 on tumor MDCs downregulates T-cell immunity and that PD-L1 blockade may represent an approach for cancer immunotherapy. Additionally, PD-L1 can provide positive costimulatory signals for innate and adaptive immunity and for protection against intracellular bacterial infection. The conversion to the (S)-stereoisomers is necessary for degradation of these substrates by peroxisomal beta-oxidation. Encoded proteins from this locus localize to both mitochondria and peroxisomes. Mutations in this gene may be associated with adult-onset sensorimotor neuropathy, pigmentary retinopathy, and adrenomyeloneuropathy due to defects in bile acid synthesis. Alternatively spliced transcript variants have been described. Read-through transcription also exists between this gene and the upstream neighboring C1QTNF3 (C1q and tumor necrosis factor related protein 3) gene.
